Overview: Wayrilz is used to treat certain people with immune thrombocytopenia (ITP). Common side effects include stomach pain, nausea, and diarrhea. Taking Wayrilz with food may help lower these side effects. Call your health care provider right away if you get an infection or have any symptoms of an infection, such as a fever or cough. Do not eat or drink grapefruit, starfruit, Seville orange, and any juices with them while taking Wayrilz.

What is Wayrilz used for?

Wayrilz (rilzabrutinib) is commonly used to treat certain people with immune thrombocytopenia (ITP).

Wayrilz may also be used for other conditions as determined by your health care provider.

How does Wayrilz work (mechanism of action)?

Immune thrombocytopenia is a condition where your body mistakenly destroys a type of cell in your body called platelets, which help your blood clot. Wayrilz targets different parts of your immune system to help stop your immune system from attacking platelets.

How should I store Wayrilz?

Wayrilz should be stored in its original packaging at room temperature, between 68 F and 77 F (20 C and 25 C). It can be exposed to temperatures between 59 F and 86 F (15 C and 30 C) for shorter periods of time, such as when transporting it. Store in a cool, dry place. Protect from light and moisture.

What are the most common side effects of Wayrilz?

The most common side effects of Wayrilz are listed below. Tell your health care provider if you have any of these side effects that bother you.

  • Diarrhea
  • Nausea
  • Headache
  • Stomach pain
  • COVID-19

What are the serious side effects of Wayrilz?

While less common, the most serious side effects of Wayrilz are described below, along with what to do if they happen.

Severe Allergic Reactions. Wayrilz may cause allergic reactions, which can be serious. Stop taking Wayrilz and get help right away if you have any of the following symptoms of a serious allergic reaction.

  • Breathing problems or wheezing
  • Racing heart
  • Fever or general ill feeling
  • Swollen lymph nodes
  • Swelling of the face, lips, mouth, tongue, or throat
  • Trouble swallowing or throat tightness
  • Itching or skin rash
  • Bumps on the skin called hives that can be red, pink, white, or brown depending on your skin tone
  • Nausea or vomiting
  • Dizziness, feeling lightheaded, or fainting
  • Stomach cramps
  • Joint pain

Infections. Wayrilz can increase your risk of infections, including serious infections that can be life-threatening. Call your health care provider right away if you have an infection or any of the following symptoms.

  • Burning or stinging when you pee 
  • Coughing, or coughing up blood
  • Fever, sweats, chills, or muscle aches
  • Shortness of breath
  • Diarrhea or stomach pain
  • Weight loss

Liver Damage. Liver damage (hepatotoxicity), which can be life-threatening, can happen when taking Wayrilz. Your health care provider may monitor your liver function before and while taking Wayrilz. Keep all appointments to have your blood checked. Call your health care provider right away if you have any of the following symptoms of liver damage.

  • Nausea or vomiting
  • Stomach or belly pain
  • Fever
  • Weakness or unusual tiredness
  • Itching
  • Loss of appetite
  • Light-colored poop
  • Dark-colored urine (pee)
  • Your skin or the whites of your eyes turning yellowish in color (also called jaundice)

Who should not use Wayrilz?

Allergies to Ingredients. People who are allergic to any of the following should not take Wayrilz.

  • Rilzabrutinib
  • Wayrilz
  • Any of the ingredients in the specific product dispensed

Your pharmacist can tell you all of the ingredients in Wayrilz.

What should I know about Wayrilz before using it?

Do not take Wayrilz unless it has been prescribed to you by a health care provider. Take it as prescribed.

Do not share Wayrilz with other people, even if they have the same condition as you. It may harm them.

Keep Wayrilz out of the reach of children.

Do not chew, cut, or crush Wayrilz. Swallow the tablets whole with a glass of water. 

Some products used for heartburn or upset stomach like antacids are available over-the-counter (OTC). Taking antacids, medicines called H2 blockers, or proton pump inhibitors (PPI) like omeprazole with Wayrilz can make it not work as well. Take Wayrilz at least two hours before taking an antacid or H2 blocker. Talk to your health care provider if you take a PPI. See the Interactions section for more information.

What should I tell my health care provider before using Wayrilz?

Tell your health care provider about all of your health conditions and any prescription or over-the-counter (OTC) medicines, vitamins/minerals, herbal products, and other supplements you are using. This will help them know if Wayrilz is right for you.

In particular, make sure that you discuss any of the following.

Current and Past Health Conditions. Tell your health care provider if you have any of the following.

  • Liver problems
  • Kidney problems

Other Medicines and Supplements. Wayrilz may interact with other medicines and supplements. Before taking Wayrilz, tell your health care provider about any prescription or over-the-counter (OTC) medicines, vitamins/minerals, herbal products, and other supplements you are using. See the Interactions section for more details.

Pregnancy. Wayrilz may cause harm to a fetus. Tell your health care provider if you are pregnant or plan to become pregnant. Your health care provider may have you take a pregnancy test before starting Wayrilz. If you can become pregnant, use effective birth control while taking Wayrilz and for at least one week after your last dose of the medicine. If you become pregnant or if you think you might be pregnant, call your health care provider right away.

Breastfeeding. Do not breastfeed during treatment with Wayrilz and for at least one week after your last dose. It is not known if Wayrilz passes into breast milk. Tell your health care provider if you are breastfeeding or plan to breastfeed.

Interactions

Does Wayrilz interact with foods or drinks?

Do not eat or drink grapefruit, starfruit, Seville oranges, and any juices that contain them while taking Wayrilz. These fruits can cause the amount of Wayrilz in your body to go up, which may increase side effects.

Wayrilz can be taken with or without food. If you have nausea, stomach pain, or diarrhea while taking Wayrilz, taking the medicine with food may help lower these side effects. 

It is unknown if drinking alcohol will affect Wayrilz. 

Does Wayrilz interact with other medicines (drug interactions)?

Always tell your health care provider about any prescription or over-the-counter (OTC) medicines, vitamins/minerals, herbal products, and other supplements you are using.

In particular, make sure that you discuss if you are using any of the following before taking Wayrilz.

  • An antacid containing calcium, aluminum, or magnesium
  • An H2 blocker, such as cimetidine (Tagamet), famotidine (Pepcid, Zantac), or nizatidine (Axid)
  • A proton pump inhibitor (PPI), such as esomeprazole (Nexium), lansoprazole (Prevacid), omeprazole (Prilosec), pantoprazole (Protonix), or rabeprazole (Aciphex)
  • Fluconazole (Diflucan), which is a medicine used for fungal infections
  • Erythromycin (Ery-Tab and others), which is a medicine used for bacterial infections
  • Efavirenz or ritonavir (Norvir), which is a medicine commonly used to treat HIV
  • Rifampin (Rifadin), which is a medicine commonly used to treat an infection called tuberculosis
  • Midazolam (Nayzilam), which is a medicine used for seizures
  • Rifabutin (Mycobutin), which is a medicine used to prevent Mycobacterium avium complex infection

Some other medicines may alter the blood levels of Wayrilz, and vice versa. Tell your health care provider about all medicines that you take or have recently taken.

Overdose/Missed Dose

What should I do if I accidentally use too much Wayrilz?

If you or someone else has used too much Wayrilz, get medical help right away, call 911, or contact a Poison Control center at 800-222-1222.

What should I do if I miss a dose of Wayrilz?

If you miss a dose, take it as soon as you remember on the same day and at least two hours apart from your next scheduled dose. Do not take double or extra doses.
